CD44 is generally regarded as one of the central markers of memory T cells who experienced exposure to antigens. However, existence of a significant frequency of CD44highCD8+ T cells has been recognized in animals, including germ free mice, who have not experienced foreign antigens. These cells have innate features like NK cells. In the present study, we observed that mouse splenic CD44highCD8+ T cells as well as NK cells, but not CD44lowCD8+T cells, expressed functional IL-18 receptors, ? and ? chains, together with IL-2 receptors CD122/CD132, CD62L, CCR7, and chemokine receptor CXCR3. In response to IL-18 stimulation, phosphorylated NF-?B, S6/p70 protein, eIF-1, BCl-XL, and ATP levels, were up-regulated together with augmented expression of CD25, Notch-related c-Kit (CD117), and PD-1 in these cells, and progressed into the G1/S phase in the cell cycle. IL-18-primed cells, expressing a high-affinity receptor for IL-2, exhibited robust expansion in response to IL-2 and underwent differentiation into effector cells with strong cytotoxicity and ability to produce cytokines. While IL-18-primed cells retained high levels of Tcf-1, the expanding cells induced by IL-2 diminished expression of Tcf-1, suggesting the decrease of stemness in them. Stimulation of CD44highCD8+ T cells with IL-18 upregulated Notch-1 receptor and c-Myc, and inclusion of ?-secretase inhibitors attenuated the effect of IL-18 on them. Notably, the effects of IL-18 and IL-2 on NK cells and CD44high CD8+ T cells were inhibited by prostaglandin E2 (PGE2) agonists. In a tumor model, administration of IL-18 increased the accumulation of CD8+ T cells in both the lymph nodes and tumors, and administration of IL-18 and EP4 antagonists significantly inhibited tumor growth. These results suggested that IL-18 promotes self-renewal and functional differentiation of CD44highCD8+T cells and NK cells in a Notch signaling-dependent manner. The action of IL-18 was suggested to be influenced by PGE2 and PGE2 antagonists. It is necessary to further examine whether the apparently antigen-inexperienced CD44highCD8+T cells primed by IL-18 play roles in anti-tumor immunity.
Cancer immunotherapies have demonstrated remarkable therapeutic potential, yet they often fall short of achieving sustained responses in numerous solid tumors. Our previous studies uncovered a mechanistic model explaining cancer relapse following immunotherapy treatments which implicates a subset of stem cell-like tumor-initiating cells that can better survive from anti-tumor immune attacks. Our new studies have now raised the intriguing possibility that these tumor-initiating cells harbor unique molecular programs that enable them to shape their immune microenvironment, rendering them more resistant to anti-tumor immunity. The aim of our study is to unveil the stem cell-specific molecular program that is responsible for establishing interaction patterns between tumor-initiating cells and immune cells, which underlie immunotherapy resistance and drive cancer relapse in squamous cell carcinomas. Using a combination of immune cell lineage tracing, single-cell RNA-sequencing and spatial transcriptome profiling, we have uncovered an intriguing dialogue between tumor-initiating stem cells and neutrophils in various forms of squamous cell carcinomas. While successful immune checkpoint blockade treatments can effectively program neutrophils, allowing majority of tumor-associated neutrophils to regain anti-tumor activities, we uncovered a subset of neutrophils residing at the tumor stroma interface that can retain their immune suppressive functions. Importantly, we further identified a group of TGF?-responsive tumor-initiating stem cells possess the capacity to block the interferon responses in neutrophils, preventing them from being reprogrammed by immunotherapies. Delving into the mechanisms., we have identified a stem cell-specific molecular circuit that enables tumor-initiating stem cells to modulate the local lipid profiles, therefore, blocking the neutrophils in their neighborhood from responding to interferons. Our findings position tumor-initiating stem cells at the heart of orchestrating the plasticity and cell state of neutrophils in cancer. Moreover, we have uncovered novel targets that can be exploited to disrupt the communication between stem cells and neutrophils, potentially preventing cancer relapse following immunotherapy.
Macrophages play a crucial role in the progression and outcome of inflammation. In this study, we show that transketolase (TKT), a key enzyme in the non-oxidative pentose phosphate pathway, was downregulated in proinflammatory macrophages and in peripheral blood mononuclear cells from sepsis patients. TKT deficiency reduced proinflammatory cytokine production in macrophages and alleviates both sepsis and metabolic dysfunction-associated fatty liver disease (MAFLD) in mice. Mechanistically, TKT deficiency led to decreased ratios of alpha-ketoglutarate (?-KG)/fumarate and ?-KG/succinate, resulting in DNA hypermethylation and reduced chromatin accessibility of proinflammatory genes. Additionally, TKT deficiency increased levels of polyunsaturated fatty acids, which inhibited the interaction between TAK1 and TAB1, thereby blocking the activation of the TAK1-NF-?B/MAPK signaling axis. Importantly, adeno-associated virus-mediated TKT knockdown in lung or liver macrophages significantly reduced sepsis and MAFLD in mice. These findings identify TKT as a crucial checkpoint that regulates metabolism, epigenetics, and signal transduction in macrophage inflammatory responses.
